ABSTRACT
NUR RASYID. Decreased Relaxation of Rabbit Corpus Cavernosus Smooth
Muscle due to Chronic Hypoxemia by SO2 as Human Erectile Dysfunction
Model. Under direction of BAMBANG PONTJO PRIOSOERYANTO, DEWI APRI
ASTUTI, WASMEN MANALU dan AKMAL TAHER.
Chronic Obstructive Pulmonary Disease (COPD) was the chronic diseases
that decrease quality of life progressively and also decrease sexual activity especially
75% of patients become erectile dysfunction. The purpose of this study is to evaluate
the effect of chronic hypoxemia to total testosterone and free testosterore level;
changing of smooth muscle and fibrous tissue in corpus cavernous; and deterioration
of contractility and relaxation using organ bath
This experimental study consisted 3 study and used 24 local rabbit (Lepus
spp.). First study was the exposure SO2 gas chronically to become COPD by evaluate
clinical symptoms, changing PaO2 level and pulmonary histopathology. Second
study was to evaluate the changes in total testosterone and free testosterone level by
radioimmunoassay technique and testicular histopathology. Third study is to evaluate
contractility and relaxation of corpus cavernosus smooth muscle and improving of
relaxation by (acetylcholine, sildenafil citrate and zaprinast) and corpus cavernosus
histopathology with Mason-Trichrom staining technique.
The result of first study showed exposure of SO2 can caused severe
hypoxemia with decreased PaO2 level < 60 mmHg, but there were no significant
differences in histopathology. In second study, chronic hypoxemia caused congestive,
edema, peritubular fibrosis and degeneration of testicular tubule cells. There was
decreasing total testosterone and free testosterone level but no significant difference.
In third study, chronic hypoxemia showed decreasing contractility and relaxation
corpus cavernosus smooth muscle due to decreasing number of smooth muscle and
increasing fibrous tissue
